Target Information
ROCK1은 세린/트레오닌 키나아제로서 세포 분화와 이동을 조절합니다. Rho/ROCK 경로는 다양한 인간 암의 진행에 관여하며, ROCK1은 새로운 Caspase-3 기질로 확인되었습니다. ROCK은 소형 GTPase Rho의 효과기로서, MLC(마이오신 라이트 체인)의 직접 인산화 및 MLC phosphatase의 인산화를 통해 세포 수축성을 증가시킵니다.
ROCK-1은 N-말단 키나아제 도메인과 C-말단 억제 도메인으로 구성되며, Rho 결합 도메인을 포함한 가변 영역으로 연결됩니다. 세포사멸 과정에서 ROCK-I은 Caspase-3에 의해 절단되어 억제 도메인이 제거되며, 지속적이고 조절되지 않은 키나아제 활성을 나타냅니다. 이 활성화는 MLC 인산화 및 막 블레빙을 유도하여 세포사멸 실행 단계 초기에 나타나는 현상을 일으킵니다.
