Target Information
Proteolytic degradation is critical to maintaining appropriate levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, and cell cycle regulation.
The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins in eukaryotic cells, while others are degraded via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome, an ATP-dependent multisubunit complex (~2.5 MDa), consists of a 20S core and one or two 19S regulatory subunits. The 19S subunits recognize ubiquitinated proteins and mediate their unfolding and translocation into the 20S core.
Defects in this pathway are linked to diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
