Target Information
TAB2 is an activator of MAP3K7/TAK1, which is required for IL-1 induced activation of NF-kappaB and MAPK8/JNK. TAB2 forms a kinase complex with TRAF6, MAP3K7, and TAB1, serving as an adaptor linking MAP3K7 and TRAF6. Together with TAB1 and MAP3K7, it participates in TNFSF11/RANKL-induced signal transduction through activation of the receptor activator of NF-kappaB (TNFRSF11A/RANK), influencing osteoclast development and function. TAB2 and TAB3 interact with Beclin-1, regulating autophagy by dissociating upon induction and binding TAK1. Overexpression inhibits autophagy, while depletion triggers it, suggesting TAB2/TAB3 act as control points for autophagy.
