Description:
The IT2.2 monoclonal antibody reacts with human CD86 (~80 kDa surface receptor, also known as B7-2). CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated via surface stimuli such as BCR complex, CD40, and cytokine receptors. It serves as a counter-receptor for T cell molecules CD28 and CD152 (CTLA-4), playing a key role in T-B crosstalk, T cell costimulation, autoantibody production, and Th2-mediated Ig production.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL reaction volume). Cell number range: 10⁵–10⁸ cells/test.

Target Information
CD86 is one of two ligands (along with CD80) for CTLA4 and CD28. It acts as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation involving CD86/CD80 can lead to tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed by antigen-presenting cells. It binds to CD28 and CTLA-4 on T cells, respectively activating or inhibiting immune responses. Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with gallbladder squamous cell carcinoma and myocarditis.
