Product Specific Information

The monoclonal antibody eBio4E3 reacts with an epitope shared by mouse CD21 (CR2) and CD35 (CR1).
CD21 and CD35 are alternatively spliced transcripts from the Cr2 gene, resulting in surface proteins of 145 and 190 kDa, respectively.
They are expressed on mature B cells, but not on thymocytes, peripheral T cells, erythrocytes, or platelets. Expression on macrophages is minimal.

Functional Roles:

• CD21 acts as a receptor for complement component C3d and Epstein-Barr virus (EBV).
• In association with CD19 and CD81, CD21 participates in B-cell activation.
• Cr2-deficient mice show impaired inflammatory and humoral immune responses.

Clone Characteristics:

• eBio4E3 and eBio8D9 do not cross-block each other, indicating different epitope binding.
• Reported application: Flow cytometric analysis of mouse splenocytes.
• Recommended usage: ≤0.5 µg/test (100 µL final volume; 10⁵–10⁸ cells/test).
• Excitation: 488 nm; Emission: 520 nm; Laser: Blue.
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD21 (Complement Receptor 2, CR2)
Binds C3 complement fragments and plays a key role in:

• Uptake and retention of immune complexes
• Survival of memory B cells
• Maintenance of humoral immune responses
  Also serves as a receptor for Epstein-Barr virus and contributes to prion targeting to follicular dendritic cells.

CD35 (Complement Receptor 1, CR1)
Encodes a membrane glycoprotein found on peripheral blood cells, glomerular podocytes, and follicular dendritic cells.
Acts as a receptor for complement components C3b and C4b, regulating complement cascade activity.
Variations in CD35 form the basis of the Knops blood group system.
