Product Specific Information

This MILAN8R monoclonal antibody recognizes human and mouse ERK1/2 (p44/p42, MAPK3/1) phosphorylated on T202/Y204. ERK1/2 are key MAPKs involved in proliferation, differentiation, motility, and survival. Activation occurs via MEK1/2-mediated phosphorylation, and downstream targets include p90RSK and Elk-1. ERK signaling dysregulation is common in cancer.

Specificity confirmed by ELISA, flow cytometry, and western blotting.
Reported applications: intracellular staining followed by flow cytometric analysis.
Tested applications: pre-titrated for intracellular staining of human PBMCs at 5 µL/test (0.5 µg). A test corresponds to 100 µL final volume, with 10⁵–10⁸ cells/test.

Staining Protocol

Use Protocol C: Two-step (Fixation/Methanol).
Protocols A and B (cytoplasmic/nuclear) are not suitable.

Fluorophore Information

PE-eFluor® 610 excitation: 488–561 nm; emission: 607 nm.
Recommended filter: 610/20 band pass (PE-Texas Red® equivalent).

Target Information

ERK1 and ERK2 regulate meiosis, mitosis, and postmitotic functions. Activation occurs via Ras-Raf-MEK-ERK signaling cascade upon growth factor stimulation. Activated ERKs phosphorylate cytoplasmic targets or nuclear transcription factors such as c-Fos and Elk-1.
