Description:
The monoclonal antibody eBio84-3C1 reacts with CD43 (also known as leukosialin, galactoglycoprotein, and sialophorin). CD43 is a sialomucin with both adhesive and anti-adhesive functions, expressed on most leukocytes except resting B lymphocytes. Upon activation, proteolytic processing decreases surface expression. CD43 is involved in activation of T cells, B cells, NK cells, and monocytes. The counter-receptor for CD43 is CD169/SIGLEC-1, expressed on macrophages.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test. A test is defined as the amount of antibody required to stain a sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It contributes to a repulsive barrier affecting cellular adhesion, but can also promote leukocyte aggregation. Through interactions with actin-binding proteins (ezrin, moesin), CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte trafficking. It modulates LFA-1 adhesiveness and signaling, and can influence tumor suppressor p53 activity. CD43 dysfunction is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
