Target Information

IRS-1 is a major substrate of the insulin receptor and is phosphorylated in response to stimulation by insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4). It undergoes phosphorylation on serine, threonine, and tyrosine residues across various tissues. Casein kinase II mediates part of the insulin-stimulated phosphorylation of IRS-1, identifying Thr 502 and Ser 99 as key phosphorylation sites. IRS-1 includes three putative 14-3-3 binding sites (Ser 270, Ser 374, Ser 641), with the Ser 270 motif located in the phosphotyrosine binding domain responsible for insulin receptor interaction.
