Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. It also exists in a soluble form (~26 kDa). Activation of FAS induces aggregation of FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and activation of caspases leading to apoptosis. FAS can activate NF-kappaB, MAPK3/ERK1, MAPK8/JNK pathways, and is involved in proliferation signaling in fibroblasts and T cells. Multiple alternatively spliced variants exist, some lacking the transmembrane domain, which may negatively regulate apoptosis. The Fas/Fas ligand system plays roles in diseases such as AIDS, hepatitis, and cancer, and is important in antiviral immune responses.
