Target Information

T cell receptors (TCRs) are essential for distinguishing foreign from self-peptides presented by MHC class II molecules, enabling effective adaptive immune responses. Recognition of self-peptides is associated with autoimmune diseases, while mutant self-peptides are linked to tumors. Superantigen engagement of TCRs can lead to toxic shock syndrome. Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and systemic lupus erythematosus patients, potentially modulating inflammatory responses.

Most human T cells express TCR alpha-beta and either CD4 or CD8 (single positive, SP). A smaller subset lacks both (double negative, DN), with increased alpha-beta DN T cells observed in autoimmune and immunodeficiency disorders. Gamma-delta T cells, primarily epithelial, exhibit limited TCR diversity and distinct antigen recognition, contributing to antitumor and immunoregulatory activity.
