Target Information
Potassium channels are widely expressed proteins that play critical roles in both excitable and non-excitable cells. The large conductance, calcium-activated potassium channel (Maxi K⁺) is unique among potassium channels as its activation is controlled by both intracellular calcium levels and membrane depolarization.
The core Maxi K⁺ channel is a homotetrameric alpha subunit containing a voltage sensor and calcium-binding domain. In vascular smooth muscle, a beta-subunit forms a 1:1 complex with the alpha subunit, reducing the calcium concentration required for channel activation by 5- to 10-fold.
Because of its regulatory role in blood flow and blood pressure, the Maxi K⁺ beta subunit (KCNMB1) is a potential therapeutic target.
