Target Information
SMADs are members of the MAD-related family of molecules. These intracellular proteins are essential components in the signaling pathways of the serine/threonine kinase receptors of the transforming growth factor beta (TGF-β) superfamily.
SMADs are classified as:
- Receptor-regulated SMADs (R-SMADs: SMAD1, 2, 5, 8, and 9)
- Common-mediator SMAD (co-SMAD: SMAD4)
- Inhibitory SMADs (I-SMADs: SMAD6 and 7)
SMAD1, 5, 8, and 9 share high sequence homology, and antibodies may recognize common regions among them. SMAD8 and SMAD9 are often used interchangeably in literature. Mutations in SMAD9 are associated with pulmonary hypertension, primary 2 (PPH2).
