Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha) is a receptor-type transmembrane glycoprotein expressed on myeloid cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
It acts as a substrate for activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.
Binding of SIRP alpha to CD47 induces phosphorylation of ITIM motifs, leading to recruitment of SHP-1 and SHP-2 phosphatases.
This interaction regulates processes such as inhibition of macrophage phagocytosis, and activation of T cells and dendritic cells.
SIRP alpha plays roles in physiological and pathological processes, including cancer, where CD47-mediated activation inhibits macrophage-mediated tumor cell destruction.
