Product Specific Information
This product is preservative-free. It is recommended to add sodium azide to avoid contamination (final concentration 0.05%–0.1%).
Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning the specific antibody DNA sequences from immunoreactive rabbits. Individual clones are screened to select the best candidates for production.
Advantages of recombinant rabbit monoclonal antibodies:
- Better specificity and sensitivity
- Lot-to-lot consistency
- Animal origin-free formulations
- Broader immunoreactivity to diverse targets due to larger rabbit immune repertoire
This antibody has specificity for Mouse SIRP alpha/CD172a.
Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha) is a receptor-type transmembrane glycoprotein expressed on myeloid-origin cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
SIRP alpha acts as a substrate for several activated tyrosine kinases, including EGFR, PDGFR, src, and insulin receptor, and is involved in the negative regulation of receptor tyrosine kinase-coupled signaling pathways.
Binding of SIRP alpha to CD47 results in phosphorylation of ITIMs within the cytoplasmic region, recruiting and activating tyrosine phosphatases SHP-1 and SHP-2. This interaction regulates processes such as:
- Inhibition of macrophage-mediated phagocytosis
- Bi-directional activation of T cells and dendritic cells
SIRP alpha also influences cellular responses to serum, growth factors, insulin, oncogenes, and hormones, playing a key role in physiological and pathological processes. Cancer cells often overexpress CD47, which activates SIRP alpha to inhibit macrophage-mediated destruction of tumor cells.
