Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) were initially discovered as adaptor proteins linking the TNF receptor superfamily to signaling pathways, acting as key regulators of cell death and stress response. TRAF proteins contain a conserved region enabling receptor and protein interactions, activating diverse signaling cascades depending on the TRAFs involved. For instance, TRAF2 and TRAF3 bind to CD40, a member of the NF receptor superfamily critical for NF-kappa-B activation in B lymphocytes. TRAF2, together with TRAF6, is also required for CD40 signaling in nonhematopoietic cells and interacts with lymphotoxin-beta receptor (LTbetaR) in association with TRAF3, cIAP1, and Smac.
