Product Specific Information

The PA1-73139 antibody reacts with diphtheria toxin.
It has been successfully used in Immunohistochemical, ELISA, Immunofluorescence, and Western blot applications.
The antibody was raised against purified diphtheria toxoid.

Target Information

Diphtheria toxin is a 58 kDa protein secreted by lysogenic strains of Corynebacterium diphtheriae.
The toxin causes diphtheria in humans by entering the cell cytoplasm and inhibiting protein synthesis.
This inhibition occurs via transfer of the ADP-ribose group of NAD to elongation factor-2 (EF-2), rendering EF-2 inactive.
The catalyzed reaction is as follows:

NAD + peptide diphthamide → nicotinamide + peptide N-(ADP-D-ribosyl)diphthamide

The crystal structure of diphtheria toxin homodimer has been determined to 2.5 Å resolution, revealing a Y-shaped molecule with three domains:

  1. Catalytic domain (fragment A): α + β fold type
  2. Transmembrane domain (TM): 9 α-helices involved in pH-triggered membrane insertion and translocation
  3. Receptor-binding domain: flattened β-barrel with jelly-roll-like topology

The TM and receptor-binding domains together constitute fragment B.
