EFNB3는 ephrin 유전자 계열의 구성원으로, 뇌 발달 및 유지에 중요한 역할을 합니다. 특히 전뇌 영역에서 높은 발현 수준을 보여 전뇌 기능에 핵심적인 역할을 할 것으로 추정됩니다. EPH 및 관련 수용체는 receptor protein-tyrosine kinase의 가장 큰 하위 계열로, 주로 신경계 발달 과정에 관여합니다. EPH 수용체는 단일 kinase 도메인과 Cys-rich 도메인 및 두 개의 fibronectin type III 반복을 포함하는 세포외 영역을 가지고 있습니다. ephrin 리간드와 수용체는 Eph Nomenclature Committee(1997)에 의해 명명되었으며, 구조와 서열 관계에 따라 ephrin-A(EFNA)와 ephrin-B(EFNB)로 구분됩니다. ephrin-A는 GPI 결합으로 막에 고정되며, ephrin-B는 막관통 단백질입니다. Eph 수용체는 이들 리간드 결합 친화성에 따라 두 그룹으로 나뉩니다.
