Target Information

Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases implicated in allergic disorders, autoimmune diseases, and cancer.
SYK translocates to the plasma membrane upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, phosphorylating downstream adaptor proteins and initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription.
Syk binds receptor assemblies via its SH2 domains to phosphorylated ITAM motifs, activated by Src-family kinases, which also phosphorylate Syk’s activation loop.
