Product Specific Information
The mature, soluble form of human TNF-alpha was expressed in Pichia pastoris (KM71H).
GenBank Accession # P01375, amino acids 77–233.
The expressed protein was N-terminal sequenced and begins with: VRSSSRTPSD.
Purity is >95% by SDS-PAGE analyses.
Activity:
Measured by cytolytic effect on mouse L-929 cells in the presence of Actinomycin D.
Use:
For in vitro applications. Perform dose-response assays to determine optimal concentration for specific experiments.
Reconstitution and Dilution:
- Store lyophilized material at 4°C.
- Reconstitute in 1 mL distilled or deionized water.
- Dilute in buffer containing carrier protein, aliquot, and store at ≤ -70°C.
- Avoid multiple freeze-thaw cycles.
- Suitable buffers: PBS with 1% BSA or neutral media containing <10% FCS.
Target Information
TNF-alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily.
It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR.
TNF-alpha regulates immune cells, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (2–5 units) but shows a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies suggest neuroprotective functions and tumor necrosis activity in mice.
TNF-alpha plays key roles in immune responses to bacterial, viral, parasitic, and fungal infections, and is implicated in autoimmune diseases, insulin resistance, and cancer.
