Target Information
Beta-arrestin-2 (ARRB2) is an adaptor protein belonging to the arrestin family, containing N- and C-arrestin domains. It plays a regulatory role in heterotrimeric G protein-coupled receptor desensitization and enhances beta2-adrenergic receptor (β2AR) mediated nuclear translocation of ERK.
ARRB2 binds phosphorylated β2ARs, impairing their ability to activate G(S) proteins. Along with AIP4, ARRB2 acts as an endosomal sorting molecule mediating CXCR4 entry into a degradative pathway. It may also participate in hormone-specific desensitization of TSH receptors.
ARRB2 is a Ca²⁺ binding protein in retinal outer segments, interacts with P-rhodopsin, and contributes to synaptic transmission in photoreceptor cells. It is widely expressed, particularly in neuronal tissues and spleen.
