Product Specific Information
For reconstitution, add 100 µL of distilled water to reach a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments using this carrier-free antibody, an additional desalting step is recommended (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 activation leads to transcription of genes such as p21waf1, inducing G1 or G2/M phase arrest. Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation.
p53 functions as a key anticancer mechanism by blocking proliferation of damaged cells. Its dysregulation is linked to aging, oxidative stress, and cognitive deficits (e.g., LTP impairment).
Mutations in the TP53 gene are common in human cancers and are also found as germline mutations in cancer-prone families such as Li-Fraumeni syndrome.
