Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, a hormone essential for reproductive functions such as establishment and maintenance of pregnancy.

PR exists in two forms:

  • Form A (94 kDa)
  • Form B (120 kDa)
    These arise from alternative translation initiation sites.

PR-B is transcriptionally active and regulates genes for endometrial maintenance, pregnancy, and ovulation inhibition.
PR-A lacks 165 amino acids at the N-terminus, exposing a 140 amino acid inhibitory domain (ID) that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription.

A null mutation in the PGR gene leads to pleiotropic reproductive abnormalities.
