Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.

PR exists as two forms:

  • Form A (94 kDa)
  • Form B (120 kDa)

These forms arise from alternative translation initiation sites. PR-B is transcriptionally active, responsible for gene activation related to endometrial maintenance, pregnancy support, and inhibition of ovulation. PR-A lacks 165 amino acids at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, conformational changes allow receptor dimerization and binding to progesterone response elements (PRE), leading to transcription activation. Null mutations in the PGR gene result in pleiotropic reproductive abnormalities.
