Product Specific Information

Description:
The 104 monoclonal antibody reacts with the mouse CD45 molecule, the leukocyte common antigen (LCA) in CD45.2-expressing mouse strains. The strains that express CD45.2 include C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising approximately 10% of lymphocyte surface proteins. It is crucial in lymphocyte development and antigen signaling, regulating Src-family kinases.
CD45.2 exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. These isoforms vary in membrane domain localization and expression depending on cell type and physiological state.
Beyond immunoreceptor signaling, CD45.2 promotes cell survival via integrin-mediated pathways and participates in DNA fragmentation during apoptosis. CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid subtypes.
