Target Information

Cytokine receptor signaling is negatively regulated by a family of Src homology 2 (SH2) domain-containing adaptor molecules termed SOCS (Suppressor of Cytokine Signaling).
There are eight members of the SOCS family: SOCS-1, 2, 3, 4, 5, 6, 7 and CIS.
SOCS proteins consist of:

• N-terminal region of variable length and amino acid composition
• Central SH2 domain
• C-terminal SOCS box motif

SOCS proteins form a classical negative feedback loop that regulates cytokine signal transduction via a STAT-induced transcriptional mechanism. Transcription of SOCS genes is rapidly induced by cytokines both in vitro and in vivo. Once produced, SOCS proteins inhibit signaling through various mechanisms.
