Target Information
SIRT5는 Sirtuins(Sir2-like proteins) 계열에 속하는 인간 단백질로, 원핵생물과 진핵생물 모두에서 발견됩니다. 모든 Sir2-like 단백질은 세균에서 인간에 이르기까지 보존된 서열 모티프를 포함하는 sirtuin core domain을 가지고 있습니다. 세균, 효모 및 포유류의 sirtuins는 NAD를 대사하며, mono-ADP-ribosyltransferase로 작용할 수 있습니다. 이 효소의 기능은 완전히 규명되지 않았지만, 최근 연구에서는 histone-활성화 Sir2 매개 NAD 대사 및 NAD-활성화 Sir2 매개 histone 탈아세틸화가 Sir2, NAD, 그리고 acetylated histone의 N epsilon-acetyl-lysine 그룹 간 상호작용에 의한 활성화 메커니즘을 가질 가능성을 시사합니다.
