Target Information
Tumor necrosis factor (TNF)-induced signaling is mediated through association of TNF receptor (TNFR) with adaptor proteins such as TNF receptor associated factors (TRAFs).
TRAFs are cytoplasmic adapter proteins that mediate signal transduction from TNF-receptor superfamily members (e.g., RANK, CD30, CD40) and interleukin-1 receptor.
The C-terminal region of TRAFs is required for self-association and receptor interaction following ligand-induced oligomerization.
Six TRAFs (TRAF1–TRAF6) have been identified. TRAF3, originally named CRAF1, interacts directly with the CD40 cytoplasmic tail through a region similar to TNF receptor-associated factors.
TRAF3 binds a single site containing the PEQET sequence, while TRAF1 and TRAF2 can bind both PEQET and an additional downstream domain.
