Target Information
RIP1은 세린-트레오닌 키나아제로, 세포사멸 수용체 결합, 병원체 인식 수용체(PRRs) 활성화, DNA 손상 후 염증 및 세포사멸 신호를 전달합니다. TNF-α에 의해 TNFR1이 활성화되면 TRADD와 TRAF2가 수용체에 모집됩니다. RIP1은 DAB2IP를 Ser-728에서 인산화하여 MAP3K5-JNK 경로를 활성화하고, TRAF2에 의한 Lys-63 연결형 유비퀴틴화는 NF-κB 및 MAPK 경로의 하류 신호 전달을 강화합니다. 유비퀴틴화된 단백질은 IKK 복합체의 조절 서브유닛 IKBKG/NEMO에 결합하여 NF-κB 활성화를 유도합니다. RIPK1과 RIPK3의 결합은 괴사 유도 복합체를 형성하여 세포사멸을 유도합니다.
