Target Information

The Ras superfamily of GTPases, which includes the Ras, Rho/Rac, Sar, Rab, ARF, and Ran subfamilies, regulates multiple cellular processes such as cytoskeletal rearrangement, nuclear signaling, and cell growth. These GTPases act as molecular switches toggling between active (GTP-bound) and inactive (GDP-bound) states. Guanine nucleotide exchange factors (GEFs) catalyze this activation.
Dbl-related proteins form a large family of structurally similar molecules that catalyze GEF activity for specific Ras family members, including FGD1, Lsc, RhoGEF p115, Lfc, Lbc, and Brx. Lsc, Lbc, and Lfc share sequence homology and exhibit exchange activity toward Rho family GTPases. RhoGEF p115 specifically catalyzes GEF activity for Rho but not Rac, Cdc42, or Ras GTPases.
