Target Information

The urokinase-type plasminogen activator receptor (uPAR) is a key molecule regulating cell-surface plasminogen activation and plays an important role in both normal and pathological processes. The human PLAUR cDNA encodes 335 amino acids, including a signal peptide of 22 residues and a hydrophobic C-terminal portion. It produces a highly glycosylated ~50 kDa protein in monocytes, anchored to the plasma membrane by glycosyl-phosphatidylinositol linkage.

Biological significance:

  • uPAR interacts with the carbohydrate-binding domain of SELL in neutrophil membranes, analogous to its interaction with β2 integrins.
  • PLAUR-mediated calcium mobilization is SELL-dependent.
  • UPAR mRNA levels correlate with the invasive potential of endometrial carcinomas, showing up to a 33-fold increase in advanced clinical stages compared to normal tissue.
  • UPAR protein expression correlates positively with recurrence rate and mortality in endometrial cancer patients.
  • UPAR serves as a useful prognostic marker for advanced endometrial cancer.
