Target Information
The Lipin family of nuclear proteins includes Lipin-1, Lipin-2, and Lipin-3, which share a nuclear signal sequence, a conserved amino-terminal (NLIP) domain, and a carboxy-terminal (CLIP) domain.
LPIN1 (Lipin-1) is essential for normal adipose tissue development and metabolism. It selectively activates PGC-1α target pathways, including fatty acid oxidation and mitochondrial oxidative phosphorylation, by inducing PPARA expression.
LPIN1 also suppresses lipogenic programs and circulating lipid levels. Overexpression promotes fat accumulation and insulin sensitivity, while deficiency can lead to lipodystrophy characterized by loss of body fat, fatty liver, hypertriglyceridemia, and insulin resistance.
