Description:
The EOS9.1 monoclonal antibody reacts with human CD95 (Fas, Apo-1), a 40–50 kDa member of the TNFR superfamily. CD95 is expressed by a broad range of hematopoietic and non-hematopoietic cells including monocytes, neutrophils, activated lymphocytes, and fibroblasts. Interaction of CD95 with its ligand (FasL) induces apoptosis and plays a role in peripheral tolerance. EOS9.1 does not block binding of DX2, another antibody specific for CD95.

Applications Tested:
EOS9.1 has been tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤1 µg per test (in 100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information
FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor within the TNFR superfamily. A soluble 26 kDa form also exists. Activation of FAS leads to aggregation of FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and caspase activation, triggering apoptosis. FAS can activate NF-κB, MAPK3/ERK1, and MAPK8/JNK pathways, contributing to proliferation signaling in fibroblasts and T cells. Multiple alternatively spliced transcript variants exist, some lacking the transmembrane domain, which may negatively regulate apoptosis. The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
