Background

This gene belongs to the protein kinase superfamily. The encoded protein contains a protein kinase-like domain but is considered inactive due to the lack of several residues required for activity. It plays a critical role in TNF-induced necroptosis via interaction with RIP3, a key signaling molecule in the necroptosis pathway. Inhibitor studies and gene knockdown have shown inhibition of TNF-induced necrosis. High levels of MLKL and RIP3 are associated with inflammatory bowel disease in children. Alternatively spliced transcript variants have been described for this gene. (RefSeq, Sep 2015)
