Specificity Comments:
The peroxiredoxin (PRX) family comprises six antioxidant proteins (PRX I–VI) that protect cells from reactive oxygen species (ROS). These proteins primarily utilize thioredoxin as the electron donor for antioxidation. PRX proteins are involved in cell proliferation, differentiation, and gene expression.
PRX I, II, IV, and VI show diffuse cytoplasmic localization, while PRX III and V are mitochondrial. Human PRX IV is expressed in many tissues and exists as a precursor (testis) and a secreted form. PRX IV is highly expressed in lung cancer and promotes tumor growth in vitro. Its expression correlates with recurrence and shorter disease-free survival in early-stage lung squamous cell carcinoma, making it a useful prognostic marker.

Target Information

Peroxiredoxin (Prx) proteins constitute a peroxidase family associated with cell proliferation, differentiation, and apoptosis. Approximately 50 isoforms exist, characterized by active cysteine residues and thiol-specific antioxidant activity.
Mammalian Prx members are grouped into typical 2-Cys, atypical 2-Cys, and 1-Cys subtypes. Except for Prx VI (1-Cys), the other five 2-Cys Prx isoforms exhibit thioredoxin-dependent peroxidase activity utilizing thioredoxin, thioredoxin reductase, and NADPH.
Mammalian Prxs (20–30 kDa) vary in subcellular localization:

  • Prx I, II, VI: Cytosol
  • Prx III: Mitochondria
  • Prx IV: Endoplasmic reticulum and secreted form
  • Prx V: Peroxisome, mitochondria, and cytosol
