This YCATE55.9 monoclonal antibody reacts with canine CD8 alpha, a cell surface receptor expressed as either a heterodimer (CD8 alpha/beta) or homodimer (CD8 alpha/alpha). CD8 alpha is found on thymocytes, subsets of mature T cells, NK cells, and intestinal intraepithelial lymphocytes. It binds to MHC class I molecules and plays roles in thymocyte development and T cell activation.

This antibody has been reported to inhibit cytotoxic T cell generation and function in vitro and to be immunosuppressive in vivo.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on normal canine peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore: PerCP-eFluor® 710 (Emission: 710 nm, Excitation: 488 nm, Blue Laser)
Recommended filters: 710/50 bandpass (preferred) or 695/40 (acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with T cell receptor (TCR) for MHC class I molecules and plays a key role in T cell activation and differentiation. CD8 is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells. The CD8 alpha chain is essential for binding to MHC-I, and its activation leads to CTL-mediated immune responses against pathogens and tumor cells. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
