Product Specific Information
MA1-861 detects Her-4 from human and mouse samples.
It has been successfully used in immunoprecipitation, immunohistochemistry, and Western blotting procedures.
By Western blot, this antibody detects a ~185 kDa protein representing Her-4 from human A431 and mouse NIH3T3/HER4 cell extracts.
It has also been used for immunohistochemistry in paraffin-embedded mouse NIH3T3 cells.
The immunizing peptide corresponds to amino acid residues 1250–1264 of human Her-4 oncoprotein.
Target Information
Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase family, including EGFR, ERBB2, and ERBB3.
ERBB4 binds and is activated by neuregulins and other factors, inducing cellular responses such as mitogenesis and differentiation.
It is a receptor for heregulin and mediates HGL-stimulated tyrosine phosphorylation of itself.
ErbB4 regulates key cellular processes including metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.
It is predominantly expressed in several breast carcinoma cell lines and in normal skeletal muscle, heart, pituitary, brain, and cerebellum.
Breast tumor cell lines T47-D, MDA-MB-453, BT-474, and H3396 show high mRNA levels, while MCF-7 and MDA-MB-361 show intermediate levels.
Expression is low or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3.
Elevated ErbB4 levels in certain human tumor cell lines suggest its potential role in malignancy.
