Target Information
VEGF (vascular endothelial growth factor)는 45 kDa의 동종이합체성, 이황화결합된 당단백질로 혈관신생(angiogenesis)에 관여하며 종양의 성장과 전이에 중요한 역할을 합니다. VEGF는 혈관내피세포의 생존, 유사분열, 화학주성 및 혈관 투과성을 촉진합니다.
VEGF 패밀리에는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, PIGF가 포함되며, VEGF 및 그 수용체 시스템은 혈관신생의 세포 신호 조절에 핵심적입니다. VEGF는 FLT1(VEGFR-1) 및 FLK1/KDR(VEGFR-2) 티로신 키나아제 수용체에 결합하며, 이들은 주로 내피세포에서 발현됩니다.
또한 FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF를 높은 친화도로 결합해 혈관신생을 억제하는 역할을 합니다. VEGF의 상승된 수준은 POEMS 증후군(Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes)과 관련이 있습니다.
