Product Specific Information

The B20.1 monoclonal antibody reacts with mouse T-cell receptor (TCR) V alpha 2 chains, recognizing the majority of the TCR V alpha 2 subfamily in mice carrying the a, b, and c haplotypes. It also reacts with V delta 8 products due to high homology.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse lymph node cells (≤ 0.1 µg/test, 100 µL final volume).
Recommended Fc receptor preblocking: 0.5 µg purified anti-mouse CD16/32 (Product # 14-0161-82).
Careful titration is advised for optimal performance.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Product # C001T06F01):

• Improved formulation over CellBlox.
• Required for optimal NovaFluor staining.
• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Conjugates:

• Based on Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
• Excitation: 552 nm; Emission: 663 nm; Laser: 561 nm (Yellow).

Note: Internal testing shows NovaFluor Yellow 660 non-specifically stains B cells in SJL mice. No non-specific staining in BALB/c or C57BL/6 strains. Other strains untested.

Target Information

T cell receptors (TCR) enable discrimination between self and foreign peptides presented by MHC class II molecules, essential for adaptive immunity.
TCR recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides are associated with tumors.
Superantigen engagement of TCRs can cause toxic shock syndrome.
Autoantibodies to TCR V beta segments are found in RA and SLE patients, potentially modulating inflammatory responses.
Most human T cells express TCR alpha-beta with CD4 or CD8; a small subset lacks both (double negative, DN).
Gamma-delta T cells, found mainly in epithelial tissues, have limited diversity and distinct antigen recognition, contributing to antitumor and immunoregulatory activity.
