Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at RT for 20 min.
Target Information
CD59 (Protectin) is a small (18–20 kDa) GPI-anchored, ubiquitously expressed inhibitor of the membrane attack complex (MAC). It acts as a key regulator that protects autologous cells from complement cascade-mediated lysis. CD59 binds to complement C8 and/or C9 during MAC assembly, preventing the incorporation of multiple C9 molecules required for pore formation.
Additionally, CD59 functions as a low-affinity ligand for human CD2, contributing to T cell costimulation and lymphocyte signal transduction.
Mutations in CD59 can lead to CD59 deficiency, characterized by hemolytic anemia, thrombosis, and cerebral infarction. Multiple alternatively spliced transcript variants have been identified.
