Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R). CD127 is a type I glycoprotein (75–80 kDa) expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation. IL-7 binding triggers Jak/STAT-mediated signaling essential for lymphocyte development and T-cell homeostasis.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). Cells co-expressing CD4 and CD25 but lacking CD127 are enriched for regulatory T cells expressing Foxp3 and exhibiting suppressive activity.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells at ≤0.06 µg/test (100 µL reaction). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Target Information

CD127 (Interleukin-7, IL-7) is a glycoprotein involved in lymphopoiesis regulation. Its receptor (IL7R) is a heterodimer of alpha and gamma chains. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling events. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells. Dysfunction is associated with severe combined immunodeficiency disorders.
