Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also for HIV-2 via gp120-mediated fusion. CXCR4 acts as the receptor for the chemokine CXCL12/SDF-1, mediating intracellular calcium signaling and MAPK1/MAPK3 activation. It also binds extracellular ubiquitin, influencing calcium and cAMP levels.

CXCR4 plays critical roles in hematopoiesis, cardiac ventricular septum formation, and vascularization of the gastrointestinal tract through regulation of endothelial branching and remodeling. In the CNS, CXCR4 mediates hippocampal neuron survival and cerebellar development. It also binds bacterial LPS, mediating inflammatory responses including TNF secretion by monocytes. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection of human target cells.
