Background
Mitochondrial DNA polymerase is a heterotrimeric enzyme composed of a homodimer of accessory subunits and a catalytic subunit. The gene encodes the catalytic subunit containing a polyglutamine tract near its N-terminus, which may be polymorphic. Mutations in this gene are associated with several disorders, including progressive external ophthalmoplegia with mitochondrial DNA deletions (PEOA1), sensory ataxic neuropathy dysarthria and ophthalmoparesis (SANDO), Alpers-Huttenlocher syndrome (AHS), and mitochondrial neurogastrointestinal encephalopathy syndrome (MNGIE). Two transcript variants encoding the same protein have been identified.
