Background
In contrast to other members of the Ras family, the KappaB-Ras subfamily lacks the conserved Gly and Gln residues at positions 13 and 65, replaced by Leu residues, making them similar to constitutively active oncogenic Ras forms. These proteins act as potent regulators of NF-kappa-B activity by preventing degradation of NF-kappa-B inhibitor beta (NFKBIB), possibly by blocking its phosphorylation and mediating cytoplasmic retention of p65/RELA. They belong to the small GTPase superfamily (Ras family, KappaB-Ras subfamily) and interact with NFKBIA and NFKBIB, forming complexes with NF-kappa-B heterodimers (p50/NFKB1 and p65/RELA). Widely expressed in various tissues.
