Background
Mammalian lens crystallins are divided into alpha, beta, and gamma families. Alpha crystallins consist of two gene products: alpha-A and alpha-B, corresponding to acidic and basic forms, respectively.
Alpha crystallins can be induced by heat shock and belong to the small heat shock protein (HSP20) family. They act as molecular chaperones, forming large soluble aggregates rather than renaturing proteins directly.
Post-translational modifications reduce their chaperone activity. These aggregates typically contain 30–40 subunits, with alpha-A and alpha-B in a 3:1 ratio.
Alpha crystallins also exhibit autokinase activity and contribute to intracellular architecture. The encoded protein is considered a moonlighting protein due to its multiple functional roles.
