Target Information

Guanylate kinase (GK) catalyzes the ATP-dependent phosphorylation of GMP into GDP, essential for recycling GMP and indirectly cGMP. In prokaryotes, lower eukaryotes, and vertebrates, GK is a conserved monomeric protein (~200 amino acids). It shows structural similarity to protein A57R (SalG2R) from Vaccinia virus. Proteins containing DHR, SH3, and GK-like domains are termed MAGUKs (membrane-associated guanylate kinase homologs), including:

  • Drosophila lethal(1)discs large-1 tumor suppressor (dlg1)
  • Mammalian tight junction protein Zo-1
  • Synaptic proteins interacting with NMDA receptor subunits (SAP90/PSD-95, CHAPSYN-110/PSD-93, SAP97/DLG1, SAP102)
  • Vertebrate 55 kD erythrocyte membrane protein (p55)
  • C. elegans protein lin-2
  • Rat protein CASK
  • Human proteins DLG2, DLG3

GK has an ATP-binding site (P-loop) in the N-terminal region, not conserved in GK-like domains of MAGUKs, though MP-binding residues are retained.
