Target Information

F-Box and WD Repeat Domain Containing 7 (FBXW7) is a member of the F-box protein family, characterized by an approximately 40 amino acid motif known as the F-box. These proteins form part of the SCF ubiquitin ligase complex (SKP1-cullin-F-box), which functions in phosphorylation-dependent ubiquitination.

F-box proteins are divided into three classes:

  • Fbws containing WD-40 domains
  • Fbls containing leucine-rich repeats
  • Fbxs containing other protein-protein interaction modules or no recognizable motifs

FBXW7 belongs to the Fbws class, containing an F-box and seven tandem WD40 repeats. It binds directly to cyclin E and may target cyclin E for ubiquitin-mediated degradation. Mutations in this gene have been found in ovarian and breast cancer cell lines, suggesting a role in cancer pathogenesis. Multiple transcript variants encoding different isoforms exist for this gene.
