Target Information
Apoptosis is related to many diseases and developmental processes. Caspase-9 plays a central role in cell death induced by various apoptosis activators. After cytochrome c is released from mitochondria, it binds to Apaf-1 to form an apoptosome, which activates procaspase-9. Activated caspase-9 subsequently cleaves and activates effector caspases (caspase-3, -6, and -7), leading to proteolytic cleavage of key proteins in apoptosis.
PHAP (putative HLA-DR-associated protein) has been identified as a regulator of mitochondrial apoptosis, facilitating apoptosome-mediated caspase-9 activation and promoting the mitochondrial apoptotic pathway. PHAP also counteracts Ras- and Myc-mediated cell transformation.
