Target Information
SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance. It connects receptors of the postsynaptic membrane, including NMDA-type and metabotropic glutamate receptors, via complexes with GKAP/PSD-95 and HOMER, and links them to the actin-based cytoskeleton.
SHANK3 contributes to dendritic spine structural organization through interactions with Arp2/3 and WAVE1 complexes and promotes F-actin clustering. By regulating actin dynamics, SHANK3 influences neuronal growth cone motility and NMDA receptor signaling. It also modulates GRIA1 exocytosis and GRM5/MGLUR5 expression and signaling, controlling AMPA and metabotropic glutamate receptor-mediated synaptic transmission and plasticity.
SHANK3 may be required at early synapse formation stages and can be inhibited by IGF1 to promote synapse maturation. (Uniprot)
