Raf-1 (c-Raf)는 MAP kinase kinase kinase (MAP3K)로서 Ras family GTPase 하류에서 직접 결합하여 기능합니다. 활성화된 Raf-1은 MEK1 및 MEK2를 인산화하여 ERK1 및 ERK2를 활성화시킵니다. 활성화된 ERK는 세포 생리 조절에 관여하며 유전자 발현, 세포 분열, 세포사멸, 분화, 이동에 중요한 역할을 합니다. Raf-1 유전자의 돌연변이는 Noonan syndrome 5 및 LEOPARD syndrome 2와 관련이 있습니다.
