Target Information
Conjugation of Nedd8 to a cullin protein, termed neddylation, is an evolutionarily conserved process that activates the cullin-RING family E3 ubiquitin ligases, leading to increased proteasomal degradation of substrate proteins.
DCN1 (Defective in cullin neddylation protein 1) is required for cellular neddylation and has five human homologues: DCNL1–5.
The DCUN1D5 gene encodes a 28 kDa DCUN1 domain-containing protein 5 with a C-terminal potentiating neddylation domain. The cellular function of DCUN1D5 is still under investigation; recent studies suggest a mono-ubiquitination-mediated mechanism regulates nuclear-cytoplasmic trafficking of hDCNL1, influencing activation of cullin-RING E3 ubiquitin ligases in specific compartments.
